3D cyclorama for digital unrolling and visualisation of deformed tubes

Abstract Colonic crypts are tubular glands that multiply through a symmetric branching process called crypt fission. During the early stages of colorectal cancer, normal fission is disturbed, leading to asymmetrical or budding. The challenging shapes budding make it difficult prepare paraffin sections for conventional histology, resulting in colonic cross with only partially visible. To study situ and three dimensions (3D), we employ X-ray micro-computed tomography image intact colons, new method developed (3D cyclorama) digitally unroll them. Here, present, verify validate our ‘3D cyclorama’ unrolls deformed tubes non-uniform thickness. It employs principles from electrostatics reform tube into series onion-like surfaces, which mapped onto planar panoramic views. This enables features extending over several layers tube’s depth, demonstrated here by two case studies: (i) microvilli human placenta (ii) 3D-printed adhesive films drug delivery. Our 3D cyclorama can provide novel insights wide spectrum applications where digital unrolling flattening necessary, including long bones, teeth roots ancient scrolls.
